If you've been following our spoilers page and or been pretty close to staying on top of the Supergirl news, this first revelation won't be too surprising to you. In talking to Entertainment Weekly, Executive Producer Andrew Kreisberg noted how we will get to meet Reign and how this villain will be unlike any villain done in the Arrowverse before. Kreisberg says:
When we meet Reign, she’s just a woman. Her name is Samantha and she’s a single mom, and she has no idea that she is Reign; she has no idea that she was the baby in the pod at the end of last season, so the journey that she takes this season is watching her realize her heritage and see how it manifests.
Andrew Kreisberg went on to discuss how the choice of bringing in Reign and telling her story from a different point of view is affecting choices across the entire Arrowverse:
The thing that everybody is struggling with on Supergirl this season is 'What does it mean to be human?' I’m not only competing with myself on Supergirl, but I’m competing with myself on all the other shows that I’ve done, and by that I mean I don’t want to repeat what I’ve done on Supergirl, nor do I want to repeat what I’ve done on Arrow and Flash. Usually on these shows you either meet the big bad at the end of episode 1 or you meet them at episode 7 or 9, and you realize that they’ve been pulling the strings the whole time, and were out there working with their multi-year plan to take down the heroes or destroy the city. When we were figuring out Reign, one thing we’ve never really seen on any of these shows is we’ve never seen the big bad become the big bad.
Additionally and possibly more exciting is this little tid-bit that came from a spoiler room also at Entertainment Weekly. When we were at Comic Con this year we asked Melissa if she was going to get a new costume and she told us that Andrew Kreisberg approached her about it but she loves her current one so much, its just too classic to let go. That said, the show looks like they found a way to get Melissa into a new suit, at least for a little bit of the time, Melissa says:
There is a different suit that I will be wearing at some point this season, but I won’t tell you when or to what capacity, It’s cool, though.
A different suit could mean a lot of things. For those of you who are thinking that Supergirl's third season is to embrace the New 52 storylines wholeheartedly as well as noticed the darker logo from the newest promo; you might be lead to believe that a Red Lantern Ring could be in Kara's future. But that is just speculation for speculations sake.

BTW, that was from the 2005-2011 run, not New 52. Or, did they do it again in New 52? I don't quite remember.

As for her being in a New 52 suit, I doubt it.

My hope is that they use recent events as a vehicle for that and not as the explanation for the ring, if that makes sense.
